Comparison of noninvasive fluorescent and bioluminescent small animal optical imaging.

Optical imaging is a modality that is cost-effective, rapid, easy to use, and can be readily applied to studying disease processes and biology in vivo. For this study, we used a green fluorescent protein (GFP)- and luciferase-expressing mouse tumor model to compare and contrast the quantitative and qualitative capabilities of a fluorescent reporter gene (GFP) and a bioluminescent reporter gene ...

Hyperspectral and multispectral bioluminescence optical tomography for small animal imaging.

For bioluminescence imaging studies in small animals, it is important to be able to accurately localize the three-dimensional (3D) distribution of the underlying bioluminescent source. The spectrum of light produced by the source that escapes the subject varies with the depth of the emission source because of the wavelength-dependence of the optical properties of tissue. Consequently, multispec...

Influence of excitation light rejection on forward model mismatch in optical tomography.

Fluorescence enhanced tomography for molecular imaging requires low background for detection and accurate image reconstruction. In this contribution, we show that excitation light leakage is responsible for elevated background and can be minimized with the use of gradient index (GRIN) lenses when using fibre optics to collect propagated fluorescence light from tissue or other biological media. ...

Small animal PET imaging.

Positron emission tomography (PET) is well established as an important research and clinical molecular imaging modality. Although the size differences between humans and rodents create formidable challenges for the application of PET imaging in small animals, advances in technology over the past several years have enabled the translation of this imaging modality to preclinical applications. In ...

Development of an optical imaging platform for functional imaging of small animals using wide-field excitation

The design and characterization of a time-resolved functional imager using a wide-field excitation scheme for small animal imaging is described. The optimal operation parameters are established based on phantom studies. The performance of the platform for functional imaging and the simultaneous 3D reconstruction of absorption and scattering coefficients is investigated in vitro.
